چکیده

Blood has been the most reliable body fluid commonly used for the diagnosis of diseases. Although there have been promising investigations for the development of novel lab-on-a-chip devices to utilize other body fluids such as urine and sweat samples in diagnosis, their stability remains a problem that limits the reliability and accuracy of readouts. Hence, accurate and quantitative separation and characterization of blood cells are still crucial. The first step in achieving high-resolution characteristics for specific cell subpopulations from the whole blood is the isolation of pure cell populations from a mixture of cell suspensions. Second, live cells need to be purified from dead cells; otherwise, dead cells might introduce biases in the measurements. In addition, the separation and characterization methods being used must preserve the genetic and phenotypic properties of the cells. Among the characterization and separation approaches, dielectrophoresis (DEP) is one of the oldest and most efficient label-free quantification methods, which directly purifies and characterizes cells using their intrinsic, physical properties. In this study, we present the dielectrophoretic separation and characterization of live and dead monocytes using 3D carbon-electrodes. Our approach successfully removed the dead monocytes while preserving the viability of the live monocytes. Therefore, when blood analyses and disease diagnosis are performed with enriched, live monocyte populations, this approach will reduce the dead-cell contamination risk and achieve more reliable and accurate test results.

Dielectrophoretic concentration and separation of live and dead bacteria in an array of insulators.

Insulator-based (electrodeless) dielectrophoresis (iDEP) is an innovative approach in which the nonuniform electric field needed to drive DEP is produced by insulators, avoiding problems associated with the use of electrodes. Live and dead Escherichia coli were concentrated and selectively released by applying stepped DC voltages across a microchannel containing an array of insulating posts etc...

The effects of suspending medium on dielectrophoretic systems for separating and sorting carbon nanotubes

The separation of two different types of multi-walled carbon nanotubes is studied in a dielectrophoresis-based microchannel system in seven different solvents as the suspending medium.  A simple model was developed to predict the behavior of the multi-walled carbon nanotubes in the above mentioned system. Then, the equations of motion for the multi-walled carbon nanotubes in that system were in...

Efficient dielectrophoretic cell enrichment using a DEP-well based system

Whilst laboratory-on-chip cell separation systems using dielectrophoresis are increasingly reported in the literature, most systems are afflicted by factors which impede “real world” performance, chief among these being cell loss (in dead spaces, attached to glass and tubing surfaces, or sedimentation from flow), and designs with large channel height-to-width ratios (large channel widths, small...

Biological cell separation using dielectrophoresis

Basic IC fabrication techniques are employed in this paper with the purpose of designing a microfluidic device with a 3D electrode arrangement to separate live and dead biological cells. This is done using the concept of dielectrophoresis, which describes the transnational motion of particles due to the application of a non-uniform electrical field. The simulations where carried out using the p...

Microdevices for dielectrophoretic flow-through cell separation - Engineering in Medicine and Biology Magazine, IEEE

M any research groups have developed dielectrophoretic (DEP)-based separation systems for a wide range of particle types, including cells, bacteria, viruses and proteins [1]–[10]. Simple twodimensional (2-D) spatial separation of a binary mixture of particles on a microelectrode array is relatively easy to demonstrate; however, the implementation of a practical flowthrough separation system is ...
